Best & Fastest way from Pompeii to Rome
Is there a direct train/bus or MUST I switch in Naples? How long should I allow for the trip?

The best and fastest way is by train via Naples Centrale. Trenitalia service to Naples Centrale is hourly from Rome Termini.
At Naples Centrale, you connect to the Circumvesuviana commuter train which departs every 30 minutes, and takes about 40 minutes to reach the Pompei Scavi Station. You must go underground for this line. If the Circumvesuviana line is on strike, you can take Ferrovie dello Stato-Trenitalia (Italian State Railways) to Pompei Citta' Station, but then you must walk at least 20 minutes to reach the Site entrance. You should plan on switching in Naples. All told, I would allow 3.5 hours or so for the trip. (An hour to get from Pompei to Naples, including waiting time for Circumvesuviana), max of 30 minutes waiting in Naples for train to Rome, and then 2 hours on an express train from Naples to Rome.

I would think renting a car would be less than 450E.

I was quoted by a driver from Rome 300E for r/t Sorrento, so Pompeii should be about that much.
