Following up on Ira's comments, your first step should be to do some research, draw up a shopping list of potential things to do, places to see. (A little research, for example, would show that Florence and Siena are part of Tuscany.) There is lots to see and do in Tuscany, and 10 days, while affording the opportunity to really enjoy the area, it will barely scratch the surface of things to see and do there. Only you and your sisters know what will be of greatest interest to you.

Both Ira and Howard are correct. There is no "best 10 days" in Italy. It takes 6 weeks to see it well, so 10 days becomes an appetizer. But, you mention April and that is a factor. I would stay central and see 3 destinations:
Rome--Florence--Tuscan hill towns
Hope this helps !

For Tuscan bases, just stick with Florence and Siena. From there you can daytrip all over the province, easily. On your daytrip list should be Lucca and Pisa, Montalcino, Cortona, Montepulciano, San Gim, and if you're a wine lover, Radda in Chianti or one of the other Chianti towns.
