Hi SunDevil,
What hotel are you staying at in Barcelona? I'm glad you chose Barcelona, because as someone else mentioned, I think it is perfect for 6 days. You will have the time to see a lot plus take a day trip or two. Also, you don't have to change hotels.

If you have the time when you return, we'd love to hear back from you and hear what you think of Barcelona. Do you plan on writing a trip report?

Congrats, SunDevil! Hope it's great and as kwoo asks above that you do a trip report!

SunDevil..
Great choice.. Barcelona is always good :-)

Coming back to the museums..
While you probably know the "big ones" (which in my view are more the ones with long queues than those you could not live without having seen) like Picasso, Miro etc there are few more or less hidden gems which can be worth while.

The two following are more geared towards temporary or traveling exhibitions so it's really hit and miss. And you should check shortly before you leave if they show something that interests you.

Caixa Forum (near Pl. Espanya)

Arts Santa Monicà (Rambles/metro Drassanes)

The new Design Museum/Center shall be completed in 2012 and should become quite an attraction (if only for its architecture)

In addition, many of the "old" museums are interesting as well (and usually there are no lines)

MUHBA, History, at Pl. Rei (near Cathedreal), with excavated Roman ruins you can walk

The Museum of Catalan History (at Port Vell)

The National Arts Museum of Catalunya

Congrats! You absolutely must see the Picasso museum, extremely interesting how his early works were quite different from the cubism with which he is usually associated. Am not really a huge admirer of Picasso's but I loved that musuem. Enjoy:)
