Is it doable to drive to Assisi from Montelpuciano as a day trip? and if so, how are the roads and what is the best route?

This is a very easy daytrip, you'll even have time to see Perugia if you want. Should take no more than an hour to drive it, even on the scenic road past Lake Trasimeno. A few years ago, we were based at La Saracina between Pienza and Montepulciano and made daytrips to Gubbio, Perugia, Assisi and even Norcia.

Sure, that is doable. I would go one way and come back the other--big roads versus small roads--see your map.
