I will be in an apt. in Rome for a week in May. Can one do a day trip to Assissi? If so what is the best way to do that?

It can be done, but it will be a long day.

Probably the best way is by train, which will take 2 hours plus each way.

There are direct trains at 07.00 (arr. 09.14) and 08.14 (arr. 10.24).

Otherwise you have to change at FOLIGNO.

Assisi station is a 10 mins bus ride from the Town.
Buses leave from outside the station (- get bus tickets from the station bar).

Yes, it is doable. Just get to Termini Station early, there are about nine daily lines (FS state-run Trenitalia; beware of strikes) that will bring you close to Assisi. I chose the Express as I traveled with my Mom and wanted to minimize logistics. It was about $24/pp. The train will stop at a station on Santa Maria degli Angeli, a town about 2 miles from Assisi. Take the bus (Line C), buses leave from outside the station and drop you very close to the Basilica of Santa Chiara. Get yourself a map of Assisi ahead of time. Very walkable town, try to do this in a good-weather day, a lot of outdoor walking. I stayed overnight (Sat to Sunday) and loved it, went to Mass at Santa Chiara, lovely memories.

As a matter of fact, if you have time, walk down into town from the train station and visit the Basilica of Santa Maria degli Angeli. The building itself houses The Porziuncola, the third church St.Francis restored. A tiny chapel, but it so well preserved. On the left side of the Basilica, you will notice a fountain with about three spouts. This is what's left (still working) from an original 26-spouts fountain The Medicis built to care for the pilgrims that were coming by the hundreds to visit the Saint Francis churches.

Do not miss Assisi, it really is a very special place.
