Looking at Van Gogh biking tour in April in the Netherlands. Would like to add a few days in Amsterdam/Brugge/Brussels. Will be traveling alone-female;art is an interest. Any suggestions for hotels/areas and places to not miss or miss.

If art is you bag by no means neglect Antwerp, which is also a very unique neat old port with an old centre
Antwerp offers not only Flemish Masters in museums but rows of Art Nouveau mansions and the fabulous Middleheim statue park, one of the worlds preeminent such outdoor parks
and the Antwerpen Centraal train station is one of the great architecturally train sheds in Europe - a work of art in intself

Antwerp also has a great maritime museum.
I recommend that you do some independent bicycling from Haarlem south through the tulip fields. You might be in time for spectacular colors in the bulb fields and a visit to Keukenhof in April. Buy a map of the bicycle paths. Bike roads are everywhere, and even have their own traffic signals.
You could bike on through the dunes to the Hague. Take in Madurodam.

cowboy Van Gogh is the name of the company organising the tour. They do a great many tours in the Netherlands. It is not clear which tour the OP is taking.
I would give Brussels a miss, and add Antwerp to your list - you can visit it on the way to or from Brugge.
You should have no problems as a single female in any of these places. Plenty of museums with all sorts of art available in Amsterdam, Haarlem, the Hague and Rotterdam.
