What are your interests? History, art, shopping? etc. Will be able to suggest if you narrow it down a bit.

However, make sure you go to Westminster Bridge for the ultimate view of the Thames: London Eye (wheel) on one side, Parliament on the other...

Almost all guidebooks offer suggested itineraries, which you can then tailor to your interests and number of days. Frommers has suggestions based on 1-5 days:

And here on Fodors, there is a "Best in 5 days" - you'll have to pare this down, obviously:

The Tower of London with all the crown jewels.

The War Museum...can't remember the correct name. Great re-enactments.

St. Paul's Church and Westminister Abbey.

Madame Tussard's...if you have never been in the States.

Harrods Food Halls.

I hate to be rude, but forget going to Mme. Tussaud's - a complete waste of time unless you have months to spend in London and are looking to just kill time. It's dreadfully touristy. There's so much else to see and experience in London. I have spent a lot of time in London as my sister lives there. Here are my recommendations:

1. Definitely the Tower of London. There's nothing else like it any where else. And be sure to see the Crown Jewels while you're there.

2. St. Paul's Cathedral.

3. Westminster Abbey and Parliament.

4. The London Eye is very good and gives you an excellent view of the city.

5. If you like modern art, the new Tate Modern is stunning.

6. The Cabinet War Rooms in Whitehall are extremely interesting. This is where Churchill ran WWII and it was left as is on VE day.

7. Go to one of the markets, Camden market or Portobello Road, for a distinctive London experience.

8. Covent Garden is lots of fun (shops and restaurants).

9. Wander through some of London's lovely parks (Regent's Park, St. James, or Hyde Park).

10. Vinopolis is supposedly very good, although I haven't been there (a wine museum with tastings and a restaurant).

11. Experience the pure city electricity on Oxford Street (the main shopping district).

Have a great time.
