Your plans in Puglia are close enough to we did that I can't find anything to change. Regarding Alberabello, you are absolutely right. It's worth a half day visit at most. As for where to sleep, anyplace on the coast near Ostuni is fine.
Can't help you regarding the drive to Puglia, because we flew into and out of Puglia.
We have been all over Italy, and in some places more than once. From this thread, I have learned that we missed one place that we should visit ASAP - LeMarche.

i was wondering about driving times on our travel days;
Do you know about how many hours in the car it may take to get from Trani to the Valle D 'Itria region if we stay near Ostuni or the inland area around Fassano? And how long to you think it will take to get from this stop to our next lay over in Leece?
Does it make sense to go from Leece to Matera on our way back up north towards Amalfi and Naples, o would you stop over in Matera after Trani and then head to Leece?
I was also wondering how your trip to Puglia compared to your trips to other regions in Italy. We loved our time in Sicily and as I am preparing for this trip it seems that this region is very similar in many ways.

It takes an hour and a half to get from Trani to Ostuni, and another hour from Ostuni to Lecce. I got those times by using Google maps and I suggest you start using them too. From personal experience, they are accurate and very helpful in understanding the geography and distances involved.
If you are driving to/from Amalfi, I would visit Matera at the beginning or at the end, that is durectly before or after Lecce. It breaks up the trip as Matera is almost exactly halfway between Lecce and Sorrento. (Lecce -> Matera, 2 hours; Matera -> Sorrento, 3 hours).
We loved Puglia. It's authentic and unspoiled. There are plenty of tourists there, but they are mostly Italian.
