On our first trip to London we took the tube from the airport to Green Park Station. LOTS of stairs.... so be prepared. Fortunately, the ever polite Brits helps with our luggage! I second the Big Bus tour to get an overview of the city.

Take a cab from Paddington to your hotel. London cabs are not expensive, and it's great fun to talk to the cabbie.

Absolutely buy your Tower of London tickets ahead. Sunday is a good day to go. We got there a few minutes before opening time, and were practically at the front of the ticketholders' queue. One of the Beefeaters entertains with jokes as you wait. When the gates open, go straight to the Crown Jewels and see them befor it gets crowded. Then walk back to the gate area and join the next Beefeater tour. After that, wander around and see whatever else you want. Allow 3 or 4 hours; it doesn't take the whole day. YOu could spend part of the afternoon at the British Museum---it's free.
