I can't help you any on Jamaica, but I can for Cancun. I recently posted a trip report for Cancun. Cancun certainly has nice beaches and is known for it's nightlife. Cancun - highrise huge hotels, lots of nightlife in large discos etc.
Jamaica- totally depends where in Jamaica, can be fairly hopping or very, very quiet but never reaches the large Cancun level, JA hotels, even the large ones, tend to be not highrise, not as large.

HHHMM, Great things about both, but Cancun is just sooo much fun. You will have many choices for nightlife and you can take the bus for fifty cents (at least it used to be)into town. It runs up and down the hotel strip and they are clean and are by every few minutes right outside your hotel. Or you can catch a cab....The whole strip is only a few miles long so everywhere is kind of close. I love La Boom, but there are many new places to go as well, they have really built alot of places next to each other so that you can leave one and into another. so you can find something that will appeal to you. The beaches are beautiful blue, you will have a great time in either !

Two great choices,
Cancun is much more developed, but does have plenty of great things to do. Does not feels like Mexico but Miami. Nightlfe is when Cancun comes to life. Watch out for springbreakers and conventions. Lots of great choices.
Negril's nightlife is reggae party on the beach, a couple of discos and/or live reggae concert.
Beaches
Cancun: The beach is always somewhat choppy however it's blue color is incredible. The best beach in the Yucatan area is Playa Norte in nearby Isla Murjeles - it's to die for. Also take a day trip to Playa Del Carmen, Cozumel or the Tulum area which has nice beaches.
Negril: The beach hands down one of the top 5 best beaches in the Caribbean (#1 in my opinion). The water is glass calm, crystal clear, shallow slope, chite sand for 7 miles long and NOT backed by highrise hotels.

Been to both. For night life it is Cancun--which is on steroids.
If you stay on the Isla Mujeres Bay side of Cancun you can get both the great beachs along with calm water.
If you decide on Jamaica we would sure recommend Negril although it is way laid back compared to Cun.
BTW--agree with the post on Isla Mujeres Norte Beach--small but totally world class with very low prices.
