There are lots of tours, easily arranged by hotel staff, from Toronto to Niagara on the Lake.
For two people, it's cheaper to rent a car; for three or four people much cheaper to rent a car.
And the car lets you pick and choose where you go and what you see.
You can make the round trip easily in one day.
It's another half hour from NotL to Niagara Falls, but due to highways, geography, etc., the return trip from Niagara Falls to Toronto, skipping NotL, is about the same 1.5 hours.

You should really allow some time to stop at the wonderful wineries on the Niagra Wine Trail as well. Inneskillin for one makes a superb ice wine. Excellent wines come out of this area and the wineries are worth a visit. Unless you do not drink then skip it.
