What are you primarily looking for in a vacation spot? (Beaches, swimming/snorkeling/diving, history, shopping, or ?

If my memory is right, the best city to live in Asia is Fukuoka of Japan, have a look and see (from ASIAWEEK).

Wow.. what a question and where should I start. Asia is so diverse and it is impossible to play favorites. From the South Asia - India is the ultimate destination. It is an experience of a lifetime. Sri Lanka is worth a visit but not sure if it is safe there with all the civil wars (sad). China and Hong Kong are also great but impossible to visit all of China unless you have a month or so travelling time. I will try to skip Hong Kong (you can get the feel walking around New York - no difference). Japan is beautiful but expensive even if you are holding the great American dollar. Still, certainly an experience.

Finally, South-East Asia - the ultimate destination.

My favorites (in order of preference)
Thailand
Indonesia
Malaysia
Philippines
Singapore

Stay a couple of days each you will come back enriched with a lifetime experience.

Have fun!
