Yes, and I've been successful in doing so most of the time, by checking in at exactly 24hrs prior to departure. [Used to be 30.]
The exception is on a very full IAH-EWR flight very recently. Those exit row seats (at least the good ones) are already taken even though I was #2 in check-in. My guess is that there were platinum elites who'd got those seats in advance, and weren't upgraded to first class because the flight was so full.

Yeah, good luck. Also, you'll see a page and have to click to acknowledge your ability to understand instructions from the FA and are able to operate/lift the doors. I paid special attention last time on a 737-500, and the overwing exit door is like 45lb.
Be sure to check the jealous looks from passengers around you, seeing that you have a exit row window seat with 10-feet of legroom and full recline.

Well, bummer I checked in exactly 24 hours before but all the exit row seats showed occupied. I guess CO elites got them already. At least I was able to switch from my original middle seat to an aisle.

Patty - You may still be able to get the exit row seats. Many elites are upgraded at the 3 Hr mark, so when you get to the airport, stop at an electronic kiosk and see if any of those seats you want open up at that time.

You can definitely check-in at a kiosk. That's the main purpose of those machines.
BTW, I just want to emphasize that all the above only apply if you're flying domestically, on a e-ticket. If you have paper ticket, or if you're flying internationally, you still have to go to a regular check-in counter.
