If you were in my situation, would you overnight in London?

Hmmm, if you and the rest of your party are good at sleeping on planes, I might just skip the overnight in London. I've done it both ways, and I arrived no more fatigued after continous flights than after the overnight in London. In fact, I slept BETTER on the London-Joburg leg when I did not have an overnight in London. Some good walk-around time in London, in fresh air would be nice...as would a massage before the flight. (If pressed for time, I would skip dinner in favor of a massage.)
The only reason to do the overnight in London is to have dinner with Kavey!

AMBIEN...at the risk of sounding like a druggie, it is the best legal drug I have ever taken on a long multi-time zone trip.
One little tiny pill and I am out for 6 hours..then like magic my eyes pop open and I feel like a new woman.
Get enough to take for the first night to two after your arrival..some times I am so tired I can not sleep.

I still felt like punky the first day in Africa after our 36 hour flying marathon but the trip home was much easier.

Uhoh...Wallybrenda will get us started. I agree. But Ambien isn't just a drug... if you are on a long flight it is a miracle in a bottle. Sleep like a baby, wake up feeling very very refreshed. BTW, I only take 1/2 of a 10mg tablet, and that works just perfectly. So try half the prescribed dose...and if that hasn't made you very sleepy in about 30 minutes or so, you can take the other half. If a half-dose works, you can hoard the other half for a future trip.
