Travel in Holland

I am arriving in Amsterdam on April 23rd in the morning and will be leaving on April 26th at night. I know is a short stay but want to get the most out of it. I plan to visit the town on the first two days and then I wanted to know if it was best to take hole day tour to go to Delft or to Rotterdam ? What do you recommend? Also should I travel by train? Are there any other touristic towns I should go instead? Thanks, Simy Jelaso
