We are going to be in Maine for only a little while on our way home from the Atlantic Provinces next June. We would like some guidance in what to see in Acadia National Park in one day. We are not hikers, but understand that some places require a little walk from the parking area. Thank you.

with just one day, I would drive the park loop road, parking at the various overlooks. Walk a little bit of the shore trail (I think it's at Otter Rocks, where it comes close to the road). Have tea and popovers at Jordan Pond tea house, and drive up to the top of Mt Cadillac, perhaps for sunset. I think that hits the top highlights and will fill a day.
Good suggestions from china_cat. It's been a while since last there, but if memory serves, "musts" for stopping along the Park Loop Road would include Sand Beach, Thunder Hole, and Otter Cliffs.
Sieur de Monts Spring and the Abbe Museum are reportedly nice as well. No idea how long a visit to either may take.
I live in Maine, and agree with the previous posters. You could do all this in one day, since you won't be hiking. The Abbe Museum is small and won't take long. It's a pity you won't have more time. If you like gardens, you might go to Thuya Gardens in Northeast Harbor. You can drive up, as they're on a hill, and park in their lot. Very close by are the Azalea Gardens, very zen and peaceful.
