We plan to book 09/01 - 09/08 Alaska cruise -- Diamond Princess. (How is this ship and itinerary anyway?)
On the last day, it arrives in Whittier at 12:30 AM, ok.
1. so, on the final night, since it arrive at 12:30am, do we have to leave the ship right away, or we can stay till the next morning? If we have to leave the ship, it's gonna be a tough night.
2. If we spend 1 or 2 days in Anchorage, what are the suggestions?

You don't have to get off the ship until the morning! Take either the Alaska Railroad or a private shuttle with tour option - the Princess express shuttles zoom by lots of stuff worth stopping at. Since you're so close, you should get to Denali - one day in Anchorage is plenty. A city tour and the Alaska Native Heritage Center are the 2 things that I put on top of the list of things for first-timers to do, but there's a fair bit to do depending on your interests. Within 100 miles there's a great deal more, including a musk ox farm, and the cheapest commercial glacier walk in Alaska.
Rent a car , stop in Girdwood for a night. Drive north of Anchorage , Talkeetna, Denali. Good roads, very easy driving.
