The best way from New Jersey is by car. Take the Garden State Pkwy to Cape May, take the ferry to Lewes, DE, drive down the peninsula and over to Chincoteague. I've never seen any rail road tracks over to the island, just a drawbridge.

I did that trip last year. Going I drove the NJ Tpk. and then through MD. and Del. (look this route up on mapquest). Coming home I took the Cape May Ferry as suggested above. The Ferry route took much longer than the land route. So, it depends on you for which one you choose. And as mentioned, you absolutely need a car once you are on Chincoteague.

Be aware that there are two sections of Assateague or you may be confused by signs. There is a section in Maryland and a section, the one near Chincoteague, in Virginia. You cannot enter in one section and drive through to the other. You have to drive around.
