Long Florida Weekend - Key West, Miami - Need advice

My wife has a long weekend at the end of September (Flying down on Wednesday evening; flying back sometime on Sunday). She wants to get away, and based on knowing her, and things she has said, these are some of her requirements:

1. Must be warm
2. Must have a beach (with room to walk)
3. Must have nightlife
4. A spa would be nice
5. She wants to visit Key West.

As I am discovering, the last one is throwing a monkey wrench into the procedings. From what I read, the beaches in the Keys are not like the long sandy beaches of other parts of Florida. Without that requirement, this wouldn't be a problem. I would say that we should just spend the entire time in the Keys, but I'm afraid that she would be disappointed in the beaches. So.... my questions are:

Are there any hotels/resorts with roomy, sandy beaches in the Keys?

In the time we have (3 full days, then a travel day) would it be possible to spend most of the time at a beach resort (close to Miami?) and also get her down to Key West to experience the nightlife. I'm afraid that we would be spending most of the vacation in a car, and not on the beach. How long does it actually take to drive from Miami to Key West?

Any advice (as to whether this is possible, where to stay, etc.) would be greatly appreciated.
