Both Boston and Philadelphia are worthy re food, culture, sightseeing, and history. I'd pick Philadelphia only because it's on the way between Washington and NYC.
Can't go wrong with either city, though.

How many days and how soon? My ideal trip would be to fly to Reagan National and stay in a hotel near a Metro stop.
See what you want in DC and then take a Northeast Regional train. You could spend a couple hours in Baltimore before going onward to Philadelphia. 30th St. station is within walking distance of Independence Hall. The next day take another Northeast Regional to Penn Station in NYC.
Do NOT take a rental car or a bus between these cities.
The latest prediction for the cherry blossoms in DC is peak during the third week of March.

Philly for food. DC for sightseeing. Boston for culture and day trips. NYC for Central Park and the Met (my personal favorite museum city is probably Boston but the Met is amazing.) all are great walking towns. Boston is probably the most interesting to walk around but it's also the most expensive. DC museums are mostly free. Natural History and the Met in NYC are pay whatever you want. I can't think of anything that is cheap in Boston. Shopping is probably best in NYC but I like boutique shopping in Boston more.
You can start in DC and do all of those if time allows- just end in Boston. If time doesn't allow, I'd do D.C., Philly, and NYC. Maybe, if you like aquariums, stop off in Baltimore for the national.

Oh, and if your title question is literal- go for Boston. I love both Philly and Boston, but the only stuff that would make me choose Philly over Boston would be food (Grand Central market) or if you're really into religious freedom or Ben Franklin or medical abnormalities (the Mutter Museum is amazing). Boston is my favorite US city hands down- fantastic art, classy hotels, very walkable.

I doubt anybody has outdone me in praising the delights of my hometown of Philly on this site, but I'll agree that both Philly & Boston are about equal in just about every facet of what makes a town a great place to visit: infrastructure, history, shopping, museums, neighborhoods, culture, shows, etc. If you've already decided on NYC and Washington -- two cities I recommend over either Philly & Boston -- then you may as well make the transportation easy by doing Washington > Philly > NYC.

You all have been so helpful. We are looking to come on the 25th from U.K. FYI.
So decided on Washington. Philly and NYC.
Was thinking of hiring a car from Washington to Philly. Would that be worth doing.

Absolutely do NOT rent a car, particularly if you are coming from the U.K. I-95 is NOT a place to learn how to drive on the "wrong" side of the road!
Also, one week is nowhere near enough time for these three cities. Pick two (at the MOST -- you could easily spend TWO weeks just in NYC!) and plan to come back for other cities.

Take the train between cities, and buy tickets SOON! The saver seats sell out.
