Family of 4 traveling from NYC to Boston in October. Hate to stay in a chain hotel - any suggestions for inns or B&B's that have rooms for a family of 4? Don't want to pay for 2 rooms and most seem to cater to couples only.

I don't know of any. Most B&B's cater to couples because their rooms have only one bed. Many have restrictions for children because they're converted private homes and do not have adequate soundproofing. And, the rates at B&B's are not the bargain they were in years past.
I was at the Madisonbeachhotel.com in June and I thought it was adorable and nicely done. Had some families there, too. Lovely location!
