Used to live in Kansas City and visited Branson several times.
Would definitely spend time in St. Louis, MO and stop at the Bass Shop nearby on the way to Branson. There are some neat B&B's just outside of St. Louis that you can find on the Net. If I get a response from friends who've been there, will respond again.
However, weather-wise, expect anything at that time of year. Ice, sleet, snow, beautiful weather with great temps...one never knows!

A good route to take could be to get to Branson via Chicago first. Then there's a highway that takes you nearly straight to St Louis (I think it's Hwy 55). On the way down you might consider Springfield Illinois -- that's where Abe Lincoln spent most of his early adult life. A tour of his house (and gravesite) may interest you.
Once in St. Louis take Highway 44, which goes southwest nearly right to Branson. 44 will take you to Springfield (Missouri this time), then there's a highway that goes directly south to Branson (about an hour from Springfield).

Zoo -- wonderful, and free admission.
Mo Botanical Garden -- yr-round beauty
Arch -- museum and ride to top (if open)
You definitely should stop at Old St. Charles, off of I-70 West, in St. Louis.
The historical "Main Street" has excellent shopping and restaurants. A must see!
