The drive is about 3 hours.
sanibel island is by far my favorite vacation spot in the whole of the united states. It's definetly worth it. The beaches are great, and check out the nature preserve on the island. We saw lots of wildlife while we were there.
Yes, it is well worth it as long as you are not just doing a weekend.
How long -- 3 hour minimum -- depends on traffic which depends on the month and the day of the month.
The directions you get on Google Maps are pretty good, if not intuitive. Ity looks like there might be more direct ways to go, but all of them have problems.
The real problem is that there is no real alternative to I-75 on this route (you have to know the back roads well), and it can get badly backed up. The Sanibel bridge also backs up, and if it is during spring training and the Red Sox are playing . . . . Well, you get the idea. Just don't expect that you can be on the beach three hours after landing, and don't book an early flight back!
