If you check the Chicago Transit Authority site (www.transitchicago.com), you'll also find a link to the Google transit site as well as GoRoo. Enter your starting and ending point and they'll list the various options that are available to you.
The closest stop is at Chicago Ave and State, about 5-6 blocks from the Knickerbocker. From where are you traveling? There are several bus routes that stop on Michigan at Delaware, about a half-block from the hotel.
If you're arriving from ORD, probably the easiest way is to exit the Blue Line at Clark/Lake and catch a cab for only a few dollars. If you'll have a multi-day transit pass and want to make full use of it, take the Blue Line to Jackson and catch the Number 151 bus at the SW corner of Jackson and Dearborn. It will stop at Michigan and Delaware. The Jackson stop has elevators and the buses are the low-floor type. Just don't block seats or the aisle with your luggage.
If you have a smart phone, you can also track buses. At the bottom of each bus stop sign is a number, text "ctabus ##" to 41411 and you'll get a message telling you when the next buses will arrive there.
