Don't want to get caught up in too much traffic going to Myrtle Beach. Would it be best to leave at 4:00 a.m. on Saturday morning or drive 1/2 way down Friday afternoon and the rest Saturday morning? Also any suggestions of routes around DC and Richmond would be appreciated.

I can't offer advice for DC and Richmond, but I'd suggest when you hit I40 you go over to Wilmington and then down Route 17 to MB.
I have only driven 95-yuck-but it's the most direct route for me. Gretchen's rec is a good one.
I can tell you that there are plenty of places to stay. Your only problem with getting there early and a 4:00pm check-in is what to do while you wait. You will need to plan for that, since (many) others will be doing the same thing.
You can avoid some of the madness of I-95 by driving US 1 until you hit I-695 and take it north of Baltimore (toward Towson) and then joining I-95 near the BWI airport. This avoids some tolls and tunnels.
Do you know where exactly you are going in Myrtle? Don't depend on your GPS to get you to the right spot.
We are staying at the Marriot Ocean Watch Villas. We were told that we could use the facilities before check-in.
That's good news.
Try to get through DC before 2 pm. 95 between DC and Richmond is busy all the time, but rush hours can be stop and go for miles. There are newish Hampton Inns (I am a Hampton fan-I have no connection to Hilton) in Wilson and Rocky Mount in NC. The beds are excellent, breakfast included and both have pools.
Here's another possible route IF you don't mind some traffic lights and lower speed limits.
I-95 south to just beyond Wilmington then Delaware 896 south to US 301 South all the way across the Potomac and then pick up I-95 south AFTER Fredericksburg. Take I-295 around Richmond...more lanes and higher speed limit.
