My wife and I have kids ranging from 8 to 1 year old. We will be flying into Tampa from 3/24-3/28 and hoping to rent a 3BR place on the beach somewhere nice. We've heard so much about Siesta Key, but after hours of searching VRBO, HomeAway and FlipKey, I cannot find any places available. Does anyone have any recommendations on another comparable beach location or available lodging in Siesta Key?

I think you are likely to have a hard time renting from VRBO for less than a week. You will wind up staying at a guest house or motel.
If the price of what you need at a hotel for four nights =/> than the price of a VRBO for a week, go ahead and rent the VRBO and leave early.
