The Hotel Plaza San Francisco is not bad, and right downtown.
To get to the cruise port, take a taxi to the bus station (about $10),
then take a bus to Valparaiso ($5), then a taxi to the cruise port
($5-10). Pullman and Turbus both have buses leaving every 15 minutes.
The journey takes about 90 minutes. I like Santiago, but you might
find Valparaiso a more interesting place to stay.

Or check out cruisecritic.com for descriptions of private transfers
(usually via the wineries) for $150 or so.
The Sheraton is nice and also on the on/off bus line for city tours. You can take the shuttle there from SCL airport for less than $18 and has great restaurants, pool and outside area along with amenities in the room.
If you would like some sightseeing along with your transportation, Leonardo Cuzmar can do this for you. You can contact him at guideschile@gmail.com
His Cell Phone: 56 9 91390672
I also used him and was very pleased. You can read my review here: www.flyertalk.com/forum/18148075-post2.html
