I can't think of anything to do in preparation for a trip, other than packing, etc.

As far as activities, are you looking for formal tours or things you can do on your own?

There are snorkeling boats, jungle tours, mountain biking, horseback riding, dinner sunset cruise of the bay, zip line, fishing charters, banana boats, parasailing, etc. etc.

You can be very busy or very laid back in Puerto Vallarta. My favorite activities are the free ones around town, like strolling The Malecon (seawall promonade), visiting the cathedral, spending a day at the local beach called Playa Los Muertos, like that.

What part of town are you staying in? That will make a different because if your resort is out in Nuevo Vallarta that is 1+ hour drive from downtown, so you'll likely spend most time at your resort. If you are staying at The Marina or Hotel Zone or in El Centro... then you'll be an easy cab ride or long walk into town.

Vallarta Adventures is a well respected activities company.

Beware of people in booths along the sidewalks or storefronts selling boat cruises or free jeep rentals... most often these are a time-share sales.

We always stay in Nuevo Vallarta and find the cab ride to be about $20 and 30 minutes from town. Most of the organized tours have pickups in Nuevo now as well. In fact, Nuevo is closer to some of them. I think everyone should go to Yelapa once, though my son would disagree as he usually gets seasick. Horseback riding is also something that we do every year.

I agree w/Suze you should wait until you arrive and usually the hotel you are staying at usually has an office for tours and etc. Have fun

If you want to do things on your own your can make your own "tours" by taking a local bus to another nearby town, going on the water taxi out to Yelapa, etc.

As Suze said -Vallarta Adventures (on the net) have great tours. Very reliable company. Caletas tour recommended. I always like the Home tour sponsored by a charitable organization --See some of the very very nice homes. Please if this is your first trip to PV don't stay in the hotel zone or Nuevo Vallarta or Marina Vallarta. You'll be too much time in the taxi. The Malecon, los Muertos Beach and Old town (zona romantica) is the real flavor of PV. Find a spot on Los Muertos Beach for the most fun. Everything is at your footsteps. In old town Go to Vista Travel on the corner of Olas Altas and Basillio Badilla -- They can arrange ALL the tours. The prices are the same wherever you buy your ticket. UNLESS you buy your ticket on the internet before you go to PV (direct from Vallarta Adventures). There is so much to do just walking the malecon, they often have activities in the outdoor stage, Watch the man paint wonderful pictures with the brush in his mouth, a lady paints plates, many artists display their art all on the Malecon. Flea markets, swinging bridges, jungle trips, ATV riding, snorkeling (caletas or marietas islands), art galleries, tile showroom, silver, fashions. So much to see. Yes, there are many people always trying to sell you something, just say "no gracias."
