Where to pick up rental car in Venice

Where to pick up rental car in Venice
Will be flying into Venice and staying a few days in July before renting a car and driving throughout Tuscany. Should we pick up the car at the Venice airport or is there a more convenient place?

Piazzale Roma, as Joan says, is the most convenient place. It's next to the train station.
But if you destination is Tuscany, then you may want to take the train to Florence and pick up a car there, as TH says.

We are driving to San Gimignano to a house we are renting with several couples. My husband thinks it may be easier to drive from the airport instead of driving from Piazzale Roma area. Guessing because never been there. The Piazzale Roma is definitely closer to our hotel which is by San Marco. Appreciate your thoughts.

Leaving from P Roma is easy--at least as easy as driving out of the airport. there's only one road out.
Sometimes the rates differ and you may want to check that.
If you leave from Piazzale Roma you only need to take a vap (or walk) to get there. Getting to the airport is more complicated and more $$$.

If you want to make life easy for you, pick up the car in the piazzale Roma. It is very easy to get to the autostrada from Venice and it will be a fairly smooth ride to San Gimignano.
Taking a train to Florence and picking up the car there sounds crazy to me. Besides the inconvenience of schlepping your bags in the train, you'll have to deal with traffic in Florence and its limited traffic area (ZTL).

Yes, P. Roma is the place to rent a car.
I'm not necessarily suggesting the train, but if you did do that, you might consider taking the train to Siena, instead of Florence, and renting and returning the care there.
There's a bit of inconvenience involved, in that you'll have to change trains in FL to get to Siena (or you could take the bus from FL to Siena). On the plus side, if you're in Tuscany, you can pick up and drop off the car in Siena and not get hit with drop-off fees that might result from renting in Venice, and you won't encounter ZTL problems in FL.
You should probably compare all the costs and time factors when making a decision. Then only you can decide what's best for you.
