I would repost this with Valletta in the title so people will be more inclined to read your question...
What are you interested in? You can either take a trip out of Valetta or say in town and sightsee there. You won't have much time. Here are some of my favorite things in Malta:
Outside of Valletta:
- Hypogeum - the underground temple and well preserved. You must purchase tickets ahead as they only allow 10 people on each tour.
- Aviation Museum with WWII planes. We had an unofficial tour from the fellow selling tickets who was a WWII pilot in Malta.
- Craft Center - lots of jewelry and glassware.

Valletta:
- St. Johns Co-cathedral (so named because it shares a bishop with the cathedral in Mdina.
- Manoel Theatre (pronounced man-well) - the third oldest theatre in Europe.
- St. Paul's Shipwreck Church
- Basilica of Our Lady of Mt. Carmel
- St. Francis of Assisi church
- Grandmaster's Palace
