What is a comfort room in a hotel?

indicates that, as least for the hotels offered on this site, the comfort room (also called a double superior) has a higher rate and is larger than a standard room. It may also face a more desirable direction (view, quieter, etc.). Otherwise, the amenities appear to be the same.
But you should ask any hotel you're contemplating what the differences between the rooms and rates in that hotel are.

Most non-English European languages either lack the concept of "comfort", as in English, or use a completely different word (possibly closer to the German 'Gemutlich')
So 'comfort', or a relative like 'confort', is often used to mean something close to 'luxurious'. In travel agent-ese this might often come down to little more than 'free chocolate on your bed', or 'just costs more'

If a hotel offers multiple levels of rooms, comfort rooms are on the lower end. For example, the Hotel Amsterdam offers 5 levels of rooms. Lowest-to-highest in price are Budget, Comfort, Smoking Comfort, Superior, and Junior Suites.
