I will be in Venice in April and have to catch a 7 a.m. flight back to the states. Can I rely on getting to the airport by 5:30 a.m. or should I take away the stress and book a hotel near the airport for my last night? I don't have my Venice hotel booked yet, so I can't say what area we'll be in. Thanks in advance for your help.

No need to book near the airport, imho.
Most North Americans in Venice have a similarly-early flight. Assuming you make a change in europe (most do), it is more like a domestic flight so the extra-early arrival times are not critical (they may not even open the airline counter until 6 or so).
It depends on exactly where you are staying, but one popular option is to take water taxi to Piazzalla Roma then a land bus to the airport. But do inquire at your hotel about what strategy is recommended.
