We are going on a Celebrity cruise the first week of May. We will be traveling from Barcelona to Nice, Florence, Venice, Rome, Naples and Croatia. I would like to know what is best and cheapest way to travel from ship to cities. I would also like to know what are the must sees for these stops.

I'm assuming that you are inquiring about traveling from the ship to the port. The ship should have tenders to take you back and forth if you have not signed up for a shore excursion.
Once you are in port, it depends on the location as to what the best and cheapest way is. In Rome, I would say taxi is the best as it will get you directly into Rome fastest; however, it is definitely not the cheapest. In Venice you can walk to a Vaporetto (water bus) station and head towards your destination, e.g. San Marco; you could also take a water taxi, but that would be much, much more expensive. In towns like Split, Dubrovnik, Hvar, Korcula, Rovinj, etc. in Croatia you can walk directly from the harbor into town. I am assuming you will be transported to Florence on a bus, since it is not a port. I have not been to Barcelona or Nice.
As for must sees, you must specify your interests -- art, architecture, history, churches, food, beautiful views, etc. Buy or check out from a library a guidebook for the areas you will visit. Look online too, e.g. at Fodors or Frommers. They have lots of good information to start with.
You will find a ton of valuable information in the Rick Steves book - Guide to Med. Cruises. He gives detailed info on most or all of the ports you mention including how to get where you really want to go. Some of your ports are walk off the ship easy do's (Nice, Venice), others like Rome and Florence have somewhat complicated and possibly expensive logistics, but fear not, Rick will tell you what to do! Besides the transportation details he also give good lists of "must sees" to help you maximize your time.
