First of all try checking for car hire on here,
http://www.arguscarhire.com/
They do the searching for you & usually come up with some good rates, as long as your pick up & drop off locations are the same. I have used the site recently for friends coming to visit us in Greece & once for myself for a visit to Lesvos.
Are you just driving direct, or planning to visit places on the Peloponnese too? It's a shorter journey to drive to Gythio & get a ferry, but as they are less frequent it depends on your travelling day/time of year as to whether this is an option,

If you do have a Peloponnese itinerary it may work out better to use the Neapoli ferry one way & the Gythio one the other. There is even one a week to/from Kalamata.
On one trip there we took the car ferry from Gythio and spent a week in lovely, very quiet Kythera, and then onto Crete from Gythio. At that time it was possible to drop off the car and fly back from the islands. It was not possible to do so on a recent trip so we opted out of taking the car and flew, picked up a car there. However, it is a lovely way to explore the Peloponnesse, and add a pretty little island. We always rent with Auto Europe.
