Will you take the route through Stuttgart? I'd stop there, and either go to the Mercedes Benz Museum or the Art Museum. If you just want to walk around it has a nice little city center.
Karlsruhe is an under rated little city. I really enjoyed their collection of art museums.
Speyer is a nice little Romanisc (sorry about sp) town.
Mainz is very attractive.
If your just looking for a cute little town, there's a lot of them.
That's about a 6 hour drive between the two. I'd look at a map and pick a town about halfway. I'd avoid the larger cities like Stuttgart and go for a smaller city (Pforzheim perhaps) and then do some googling on the city name and "restaurant" get some ideas of what may interest you both from a food and walk around perspective. A restaurant by a river or park to facilitate a walk would be nice.
Heidelberg, Würzburg, Aschaffenburg or Seligenstadt might be some options too.
