There is no one website which lists everything, but there are several good ones which together will give you an idea of what's on. If you do a search here for "london theater" you will find many, many threads on the subject with excellent information.
I generally check lastminute.com about a month before I leave to see if they have special deals on anything of interest (got reserved, half-price tix to the Royal Ballet once) and then pick up a copy of Time Out once I arrive. And I also take advantage of the Half Price Ticket booth, which invariably has something I'd like to see.

I agree with the "officiallondontheatre" website as a good place to start. You can enter the dates you will be in London and it will tell you what shows are booking for then. Of course, the list may change (particularly expand) between now and then.

I use www.albemarle-london.com to get info on London shows. I usually buy tickets through the Ticketmaster link mentioned by Karl because Ticketmaster tells you the exact seat(s) you are buying.
Another good web site is www.theatremonkey.com because they tell you the best & worst seats in each theatre.

I've never understood buying theatre tickets through Ticketmaster for London, because I don't have extra money to throw away. You can call any theatre in London direct and they also will tell you the exact seats you will get. They will take a credit card by phone and hold the tickets at the box office for you until performance. The best part of this is there is absolutely NO extra fee, unlike Ticketmaster.
I do agree that Albemarle is great for the seating charts to print off before you call the box office, so you know exactly what they are talking about. And if unclear, theatre monkey will tell you even more than you ever wanted to know about the best or worst seats in any theatre.

Ticketmaster UK only charges a 1.5 pound service charge. Sometimes that service charge is actually less than the cost of the telephone call.
I've had excellent experiences with Ticketmaster UK. Tickets are held at the box office, but don't bother to pick them up until about 90 minutes prior to the show.
