We are planning a trip to Italy in May. We plan to land in Milan, travel to Florence and then travel to Venice to return home. We'd appreciate suggestions for how long to stay in each city, routes to take (we plan to rent a car), and sites to see.

Suggestions for what to see and routes to take will depend on your personal interests and the length of your trip. Are you into food, art, architecture, etc? A good guide book will tell you what is there. You might want a car only for time in the countryside. You can't drive in Venice and it is difficult in Florence. Unless you plan to sight-see in Milan, it might be faster/easier to land in Venice and depart from Florence, Pisa or Rome.
How many days do you have? I think a car for these places is a liability. You can't drive in Florence or Venice. I would use the trains.
