What is the best way to travel from Reykjavik to Akureyri to Hofn without renting a car? Are one way flights affordable? Or is a bus the only option? Thanks

There are flights for these routes. If they are affordable or not - is a matter of opinion. Air Iceland (not Icelandair!) is the airline that performs those flight. You can get all the information on their site: http://www.airiceland.is/
Many years ago we flew RT from Reyk to Akureyri. If you don't have a lot of time to spare, that is the way to go. As eri says, whether it is worth it depends on your opinion (and time and budget.)
