Hauptbahnhof to Tegel?
We will be getting into the Hauptbahnhof from Prague and want to stay at a hotel overnight before flying home to U.S. Can we leave from Hauptbahnhof by train and go directly to the area by Tegel for a hotel? Alternately, if we stayed in the city overnight, would we take the S-Bahn , U-Bahn or a taxi to the airport and is there a station in or by Hauptbahnhof. We are wanting the most stress free choice. We have to be at airport by 7:30 or so.

Do you know how to do a google search? You will quickly be lead to http://www.berlin-airport.de/en/trav...port/index.php showing your public transit options.
There no U/S-Bahn to the airport. Look at the route network map link on the page noted. While there are stations closer to the Tegel than the Hauptbahnhof, you access to transit, such as TXL bus is greater from the Hauptbahnhof or other major stops served by TXL and other buses. If you want "stress free", just arrange a taxi the night before. My flight home from Berlin is usually around 6am and it is about 25 min taxi ride at that time from pretty much any part of town with no traffic.
To find out the bus schedule, use the www.vbb.de link listed on the airport page.

I stayed at the Hotel Melia in the Friedrichstraße and took the TXL bus from Tegel to the hotel. The bus did stop at the HBH.
I took a taxi to Tegel when I left Berlin.
The taxi was faster and easier. I think it was only 15-20 minutes to the airport from Hotel Melia (down street from Museum Island).
The Friedricstraße train station was across the street from my hotel.
I don't know if you can transfer from Friedrichstraße train station to HBH. They are very close to one another.

First, you need a hotel.
I understand, you will be coming from Praha and have an evening in Berlin. If you would like to go out for a stroll in the evening, I would second the suggestion of staying in the Friedrichstraße area.
There are trains (S-Bahn) and RE from Hauptbahnhof to Friedrichstraße train station every few minutes, so it is very quick and easy to reach a hotel in the Friedrichstraße.
There are three hotels very close to the station: Maritim Pro Arte (the best one, with a pool), Eurostars and Melia. All three are in easy walking distance, even with trolleys (about 100 yards). It is about 300 yards to the Westin Grand, a classic grand hotel. Check www.hrs.de or booking.com for rates.
From these four hotels, you can do a short stroll to Unter den Linden Boulevard and to Gendarmenmarkt for pretty impressions of historical Berlin and many places to grab a bite. In the evening, this area is beautifully illuminated.
For the next morning, I would strongly advise to take a taxi, especially if you want it stress-free. The fare would be slightly less than € 20.00 and driving time 20 minutes maximum. The taxi drops you off right in front of your check-in counter (this is a specialty of Tegel Airport!).
Alternatively, you can take the Bus TXL which stops at the crossing Unter den Linden/Friedrichstraße (driving time 31 minutes),but why the hassle to save € 14?
The alternative would be taking a taxi from the Hauptbahnhof to the Mercure Airport Hotel directly at Tegel Airport. But it is a mediocre hotel in the middle of nowhere, you cannot do anything in the evening and in the morning, you will have a walk of at least 10, if not 15 minutes with baggage from the hotel to the check-in counter. So really no gain compared to a taxi from the city centre that drops you off right in front of the check-in counter.
The area around Hauptbahnhof is barren and not a good place to stay overnight.

When we are in Berlin we almost always stay in a hotel in the Potsdamer Platz area and since we inevitably leave from Tegel we simply get a taxi. It is always "the easiest" because it comes to the door of the hotel and there is nothing "easier" than that IMO.

If you are arriving Hbf, you can get the S-Bahn from Hbf to S-Savignyplatz or S-Charlottenburg - - Savignyplatz is a particularly nice option for late strolling/restaurants/bars.
These stops are a bit more toward the airport, and they are fairly low stress because they are easy access to hotels and are less-trafficked areas than Friedrichstrasse/Potsdamer Platz.
From Savignyplatz it's a 17 minute cab ride to Tegel. From S-Charlottenburg it's 15 minutes either by taxi - - or by the #109 bus which goes from Kantstrasse straight north to the airport.

For the last 20 years, I have been to Berlin at least a dozen times per year.
Savignyplatz is a trendy neighbourhood for West-Berliners to hang out (a friend of mine, an artist, lives there), but it has not the historical grandeur of the buildings in Berlin-Mitte around Unter den Linden/Gendarmenmarkt near Friedrichstraße. So, for an evening stroll, Mitte (the historic centre) is certainly the better option, even if you have been to Berlin.
I cannot confirm that the area is less trafficked and that the drive to the airport is faster. From the Friedrichstraße area, the taxi drives the northern route to Tegel. I have never experienced this route congested, especially not in the morning.
So, driving time is about the same as from the Savignyplatz. Charlottenburg would be for me the least option, compared to historic Mitte and lively Savignyplatz.
