Hamburg airport to Kiel port
My cruise leaving from Kiel in August at 4p.m Sunday and my flight from Chicago landing in Hamburg at 9:30a.m the same day.
What would be the best way to get to port of Kiel from Hamburg airport?
I would prefer a shuttle bus. I already checked the private one and it’s to expansive.

I'd take a train myself. I think that's the easiest way and probably also the cheapest and fastest. It's well over an hour to Kiel by road from Hamburg, so I wouldn't expect a private transfer to cost less than 150 euros.
But my primary question is have you inquired with your cruise line about a bus transfer? Every cruise line offers transfers from nearby airports on the day of embarkation. These are often a bit too expensive, but they are definitely convenient. And virtually every cruise line offers them even to passengers who don't book their air travel through the cruise line. I'd explore that option first if you haven't already. But expect to pay no less than $60 or $70 for this (per person).

I'd second Doug_Stallings. The train is 1hr 15mins by InterCity Express train... German trains are fast efficient and inexpensive. Searching next week it's about £25 for a one-way ticket to Kiel.
To get to Kiel train station (Bahnhof) from the cruise port, it's a 30 minute walk along Kiel Fjord. If you have a lot of baggage you could take bus no. 41, or ferry line F1 from the dock to the station.

There is also direct busses from Hamburg airport to Kiel that will take you either to the central bus terminal in Kiel (Kielius), or deliver you to any chosen destination ("house door" service; Kiel Exx):

The Kiel Exx is a door-to-airport or v.v. shuttle service and costs €29 pP. Their vans will take you directly to the cruise port (but maybe stop on the way elsewhere to drop off other passengers). These shuttles run every 30 minutes.
You should (but don't have to) reserve ahead.

The Kielius is a regular big bus that only goes to Kiel central bus station for €19. But then you have to take a taxi to the port from there.
