First time 2-week Italy Itinerary: How many cities can we cover?

My wife and I will be 28 when we go to Italy next year, and we're trying to plan the best Itinerary blending leisure with exploration. We're looking at 2 weeks starting June 1st, and think we'd like to hit the following places in this order:
- Venice (fly in)
- Lake Como
- Cinque Terra
- Florence/Tuscana
- Amalfi
- Rome (fly out)

We are very active, and enjoy hiking and exploring the beautiful places. We also really enjoy finding the best places to dine. I would order our 3 priorities like this:
1. Landscape beauty and exploration
2. Culture and food
3. History and museums

We don't want to miss the "Must see's", but we also know that realistically 2 weeks in Italy won't cover it all. We're thinking that we should choose one between Cinque Terra, Como, and Amalfi. For first-timers, what would you prioritize? What can we not miss? What should we wait to see on another trip? Which locations would you rent a car, and where would you use public transport?
