You can check with Mappy.com to see the expected drive time and distance.
I would expect to spend the night somewhere along the way since it is a fairly long drive and the actual time depends on when you're planning this trip; time of the week, time of year, etc.

It depends on how fast you drive - and how you get around Madrid.
Mappy says 5 hours from Granada to Madrid - but we did it in a little over 4 - and that wasn;t rushing. Mappy says 3 hours from Madrid to Burgos - haven;t driven that part so can;t comment.
So obviously you could do easily in one day - esp if you have more than one driver. It depends on you plans - the obvious choice is to spend a couple of days in Madrid - but you may have done already - or not have time.
