How? Car? Train? Bus?
By car the obvious answer would be a day trip - since it's only about 80 miles. You would never have time to get to Assisi, see anything and get to Naples in time to take the last ferry to Capri (they don;t run very late).
By train - or bus - I would assume the answer to be the same - but don;t have info on cross counry travel - sometimes it's a case of you can;t get there from here. You would need to check detailed train or bus schedules. It may not be possible in a day via public transport.

Hi Season, when you ask about a daytrip to Assisi from your base in Siena are you referring to travelling from Siena to Assisi and then back to Siena the same day? That is the way I read your question but I could be wrong of course. Anyway, if I understand you correctly trying to visit Assisi from Siena and returning to your base in Siena on the same day is not something you would want to try IMO.
Now about stopping in Assisi while on your way to Capri could be a good idea. But like nytraveler my question would be to ask what type of transportation will you be using. I assume you will be travelling by car but I learned a long time ago to never assume anything. And how much time to you plan on for travelling from Siena to Capri - guess I am asking not only how long do you plan for this part of your trip but where do you plan on arriving on the mainland of southern Italy to get to Capri? Best wishes.

By itself, Assisi is easily worth a day (perhaps even more). As the previous posters have noted, to drive from Siena and back in one day or stop en route to Capri on the same day, is a bit much and really doesn't allow sufficient time to enjoy the many pleasures of Assisi.

Assisi is lovely and definitely worth staying in for at least a night if possible - it is packed with tourists during the day but in the evening the peace and authenticity of the town will charm you. Hike up to the old fort before sunset for an unforgettable view of the Umbrian countryside.

Oh azure, sunset time - looking at the Umbrian countryside is something I will never forget! However in a way I did if that makes sense. It was so beautiful that particular evening that words could never describe it. Thanks for bringing back a fantastic memory.

Thanks all for your input. Sorry, forgot to mention we will have a car for that part. The itinerary we have booked our hotels for is 3 nights in Venice, train to Florence, meet another couple at the airport, drive to San Gimigiano for 2 nights, 2 nights Siena, 2 night on Capri, three nights Rome. The idea of Assisi has come up in our group of four as an afterthought after some reading about it. We visited Florence before and loved it. Will have to weigh hotel hopping, (trading a night in Siena for a night in Assisi), against a very squeezed day trip, or hope for another trip to Italy someday. Any votes?
Thanks so much for sharing your knowledge.

Wow. That is a lot of locations for 12 nights. How about skipping Capri and consolidating Tuscany into one base area from which you can see Siena, San Gimignano and maybe a visit to Assisi if its feasible. It will save lots of time going from hotel to hotel...

If you are "sold" on your current schedule, I'd say skip Assisi. There's plenty to see closer by. I love and recommend Assisi, but it will take you a solid 2 hrs to get there (not all that pretty a drive either).
If you want advice about your overall plan, I'd agree that Capri just doesn't fit your avaiable time very well. I assume you are returning the car and training there?
Do you plan to stay in Siena? No car needed there as well.
With all your checking in and out and with the great distance you are covering, I'd reconsider your destinations.
