Hi Dianne. You might want to have a look at www.visitmalta.com for some ideas, but I would suggest the following:
Take a carrozin (carriage) ride around the city walls, but agree the price before you get in.
Visit St John's Co-cathedral - absolutely gorgeous interior.
Visit The Malta Experience, an audio visual show covering Malta's long & busy history.
Sit at a cafe table on Republic Square, & watch the world go by.
Take a harbour cruise - Valetta is stunning when seen from this angle.
Upper Barraka Gardens are also worth a visit, the views from there are superb.

Definitely the cathedral. I'm an atheist and I was impressed.
The archaeological museum was also amazing--though better if you combine it with a visit to one of the temples somehow (depends on how you allot your time the rest of the day). Perhaps there are tours of some sort if you don't want to deal with the bus in your limited time. (I'd look into a tour to Medina also) An ideal tour outside of Valletta would be to hit the temples, stopping for a photo session at the cave on the southern coast, then Medina. However, there is plenty to do in Valletta to pass a leisurely day.
The other highlight for me was just walking the side streets of Valletta.
