If you're planning to hike between all five villages (if the trails are all open when you are there - some may be closed), you need at least a full day there so two nights. A third night will give you more time to slow down and relax, dwell in the villages a little without rushing through.
I guess if you arrive early or leave very late, you could do a rushed one-day hike between villages, but that would be a long day.

The higher trails are open, at least they were when I went in mid June.
The lower trails are closed and will be for at least a couple more years because of the landslides from 2011.
I did a hike from Manarola up to Volastra and down to Corniglia. Not sure how much elevation but it took 3-3.5 hours, early in the morning before it got hot.
It would have been tough to do more hiking on the same day. So if you want to hike between all the villages, you're probably talking about 1 or 2 villages at the most per day.
It took 2 days for me to visit all the villages just by train, which sometimes run only once per hour.
If you add a full day trip to Portovenere and Palmaria by boat (and longer to take the train to La Spezia and then take the boat or bus to Portovenere) and maybe a day trip to Santa Margherita Ligure and Portofino, then you're at 4 full days, 5 nights.
You can get off at each train station, walk around the center of the villages, maybe walk to the marina, get lunch, you can probably do it in two full days. But hike above the villages and those will eat up time and energy.
However, the time spent is totally worth it if the weather is clear.

If you get rain, the rangers may close the trails (even those that are generally open). So if hiking the trails is a high priority, you might want to plan on an extra night to give you at least some latitude in the event of bad weather.
