Car Rentals in Avignon
For a trip I'm planning for next spring, we will be renting a car in Avignon for a week in Provence and then traveling to Nice. I know there are excellent train options to the Riviera, but I'd be interested in recommendations for one-way car rentals out of Avignon.

Just book whatever is cheapest in the category you want, I'd say, from the location you want (whether in-town or the TGV station). I don't see what difference it makes if it's a regular large name car rental company.

I always use www.kemwel.com . Same company as AutoEurope - but usually better rates in France than AutoEurope.

As for routings IMO avoid the busy coastal roads and go via Aix-en-Provence and then continue inland via Moustiers and the Grand Canyon de Verdun - western Europe's biggest canyon - and over to the Route de Napoleon -built for military reasons by Napoleon - down into Grasse and Cannes. The hinterlands of eastern Provence.
