You did not tell what do you prefer to visit.
I would select between two possibilities:
1. Stop in Fussen (to visit Ludwig's palaces and Oberamergau)
2. Stop in Meersburg (on Constance Lake) to visit the small towns around lake and Mainau Island.
Thanks so much! We are up for anything - open to all recommendations. Will likely stay overnight, one night.
Depending on your driving route you might consider Ulm ("the gold of Ulm rules the world") or Augsburg although the latter isn't really that far from Munich.
You could go "out of your way" a bit to see Nurnberg but IMO that city deserves more than a simple overnight.
I would suggest the university town of Tubingen, much less touristy than its regional rival Heidelberg. You could stay at the Hotel am Schloss
