I am travelling with my friend. We are young (20) and are thinking of taking this route around Spain: Barcelona, San Sebastian, Salamanca, Seville, Valencia, Madrid. Is this the best option? Are there any other places you could reccommend? We are also open to any suggestions of best ways to get around.

You have a choice of three options, bus, train and in some cases flying. The last option is car a hire but if you are only visiting cities then it is considered a waste of money.
But to answer your question we need further explanation of what do you mean best?
How long have you got for this trip? And when is this trip planned for?
To answer your question about other suggestion we need to know what interests you.
I'd do it by train but with the option of air for any jumps. It depends on what you want to do but I'd have a look at

You might like the Picos d'europa a strange mountain area, with good walking, still relatively unexplored, some very old churches and a large part of the true cross.
I hope you have at least a month. I would fly into Madrid and head to Seville,Cordoba, Cadiz, Ronda, Malaga, Granada, fly to Barcelona (or train), train or fly to San Sebastian, Burgos, Salamanca and back to Madrid. If you have time I would include Santander, Picos de Europa and Santiago de la Compostela.
