Best location for hotel in Milan for 2 nights

What do you want to see or do while you're in Milan? Two nights is a very short time for such a big city, so you should stay near whatever spots you want to visit.
I've never stayed at the Hotel Berna, but I've walked past it.

The Berna seems to consistently get good reviews. When we were last in Milan we stayed in a hotel located closer to LaScala because that was the main reason we were in the city. Therefore I agree with Bvlenci's recommendation.

Thanks. I booked a day tour that includes the Last Supper but the tour meets at Milan visitor Center at Large Cairoli. Would it be better to stay close to the meeting place since it's such a short visit?

We stayed at the Hotel Star. It's maybe 500 meters from the Duomo and even closer to La Scala. I recommend it.
Maybe I'm missing something, but you ask "Is it better to stay near the Duomo or by the Central area?" Isn't the Duomo near the "Central Area"?

We've also stayed at the Hotel Star. Multiple times, actually, although not very recently. It's within walking distance of Cairoli and very close to the Duomo, Galleria and La Scala. A great location for a short stay, IMO, but the Star lacks the charm that some people might require.

That's what I assumed.
Largo Cairoli is an easy walk from the Duomo, so I'd stay near there. We've stayed in the Hotel Ariston and the Ambasciatori, both in that area (although the Ariston is farther away). However, I always just check what kind of bargain I can find on www.booking.com . You can enter your destination as "Duomo di Milano" and it will show you hotels in that area. You can sort the list by price or by guest rating. Be sure to read the guest reviews before making a decision.
