The Marriott is NOT that near to the Acropolis, it's a couple of kms away!!
The X95 gets you to Syntagma Square, runs 24/7, costs €3.20. From there you could grab a taxi to the hotel. There are buses that run close, but with luggage I would do the taxi for that last bit. Once you are there you have the metro and tram stops at Syngrou/Fix about 1 km away or a bus stop just up the road, for exploring the city.
A taxi from the airport direct to the hotel would be around €40 but can vary depending on traffic.
This site gives you some idea of location & distances, although it does state 4km to Acropolis & Plaka! I would say closer to 2km & Google Earth confirms this!
